Immunoprecipitation

Immunoprecipitation is a highly selective technique which can be used to enrich or concentrate low abundance species from a complex matrix. Immunoprecipitation is a common first step for experiments such as selectively enriching for phosphoproteins or peptides for post-translation modification studies or concentrating a biomarker prior to quantitation. You do not need to elute from your IP beads to use the PreOmics kits, just wash the beads (without blocking proteins or detergents) and then begin! This minimises losses and speeds up processing.

The iST/iST-NHS kits are suitable for use in research and regulated environments. The kits contain all you need for your peptide or protein sample preparation and are suitable for use by all members of your lab, no specialist experience is needed. The flexible formats of single samples or racked in to 96 well holders allow the kits to be used manually or with a range of automation options. The cartridges can be processed by centrifugation or using positive pressure manifolds, again giving options for all labs. Dedicated resources are shown below and our technical support team would be happy to help with advice.

Non-depeted plasma/serum samples:

Final wash of the IP beads should not contain blocking protein e.g. BSA or detergents. The beads can be transferred to the cartridge or processed in a microcentrifuge tube for the PreOmics methodology.

Agarose bead immunoprecipitation:  

Final wash of the IP beads should not contain blocking protein e.g. BSA or detergents. The agarose beads should be proceeded in microcentrifuge tube for the LYSE and DIGEST steps, then transfer the resulting peptide mixture to the cartridges for the clean-up step.

Coated well plate immunoprecipitation:

Final wash of the IP beads should not contain blocking protein e.g. BSA or detergents. The PreOmics reagents can be added directly to the wells and mixed well for the LYSE and DIGEST steps, then transfer the resulting peptide mixture to the cartridges for the clean-up step.
