Simplifying mammalian tissue homogenization with BeatBox®
Among the biggest challenges when preparing fresh or frozen mammalian tissue samples for proteomic analysis are the tissue composition variability and properties of the samples. Tissues can vary significantly in protein amount, hardness, elasticity, and fibrous content, often making it difficult to develop a universal protein extraction method.
To disrupt extracellular structures, mechanical homogenization methods must be employed, and conventional methods are usually time-consuming, laborious, and low throughput, which can be a limiting factor when processing large numbers of samples.
Achieving efficient protein extraction is key to obtaining an unbiased representation of the proteome. Especially when working with small tissue pieces such as needle biopsies, it can be challenging to get sufficient protein content due to the risk of sample loss. On the other hand, multiple proteomic assays require larger input amounts of mammalian tissue, and scientists need a homogenization method that delivers efficient protein extraction for a broad range of projects.
The BeatBox® is a fast and easy solution for homogenizing up to 96 fresh or frozen mammalian tissue samples. It addresses all the challenges mentioned above by offering a simple yet flexible homogenization workflow that is not only compatible with different tissue rigidities but also with various input amounts. Poster
The BeatBox® Tissue Kit 96x provides a reliable solution for high-throughput analysis of low-input samples, such as for biomarker discovery from needle biopsies. Check out this poster to learn how scientists from the Matthias Mann lab (MPI of Biochemistry) are enhancing protein extraction for LC-MS analysis, starting with as little as 1 mg of various mammalian tissue types in 96-well format.

Technical Note
For proteomic applications requiring larger starting amounts, such as the analysis of post-translational modifications (PTMs), the BeatBox® Tissue Kit 24x offers the flexibility to process up to 50 mg of tissue in 24 single tubes. In this technical note, BeatBox homogenization is compared with conventional bead-beating technology for LC-MS analysis of various mammalian tissue types. Discover how the BeatBox® can simplify and reliably improve your protein yield and proteome coverage for tissue input amounts ranging from 5-50 mg.
