Optimizing plasma and serum sample preparation for deeper, high-throughput proteome profiling

Unlock deeper biofluid proteomics for biomarker discovery – From complete, ready-to-use sample preparation kits to dedicated CRO services

Flexible and scalable solutions for plasma, serum, CSF, and other complex biofluids, enabling reproducible low-abundance protein detection for biomarker discovery and translational research.

The challenge

Biofluid proteomics requires both depth and reproducibility

Plasma, serum, CSF, and other complex biofluids contain proteins across an exceptionally wide concentration range. In LC-MS-based proteomics, signals from highly abundant proteins can mask biologically relevant low-abundance proteins - a challenge that becomes even greater in large-cohort studies requiring reproducible and scalable sample preparation. Addressing both aspects is essential for achieving deeper proteome coverage and revealing meaningful biological insights at scale.

The solution

Comprehensive biofluid proteomics workflows from sample to insight

Enrichment technologies provide access to a broader range of proteins within complex biofluids, increasing the visibility of biologically relevant low-abundance proteins. Integrated with the standardized iST technology, enrichment becomes part of a streamlined end-to-end process that combines proteome depth with reproducibility, scalability, and automation readiness. This enables confident analysis across diverse applications, from exploratory research involving complex, high-dynamic-range sample matrices to large-scale cohort studies. Together, ENRICH-iST and P2-iST Plasma give researchers the flexibility to choose the solution that best matches their study objectives, whether the goal is robust large-scale profiling or deeper exploration of the biofluid proteome.

ENRICH-iST – Biofluid sample preparation simplified

ENRICH-iST is a fast, flexible, antibody-free workflow for deeper proteome profiling across plasma1, serum2, CSF3,4,5, saliva6, cell culture media7, and other complex biofluids. Using single paramagnetic bead-based enrichment, it combines enrichment, digestion, and peptide purification to deliver LC-MS-ready peptides from low sample volumes within hours. Compatible with human and non-human mammalian8,9,10 samples, ENRICH-iST supports reproducible, scalable, and automation-ready biomarker and discovery proteomics.

Deeper proteome coverage compared to antibody-based depletion

Deeper proteome coverage from minimal sample input

Unlock deeper plasma and serum proteome coverage from just 20 µL of sample input. ENRICH-iST is species- and matrix-independent, achieves proteome depth comparable to depletion strategies, provides complementary biological information, and delivers approximately 2-fold deeper proteome coverage than neat sample preparation workflows.
Streamlined workflow for validation of known and novel biomarkers

Complementary insights beyond affinity-based proteomics

ENRICH-iST provides deep, unbiased proteome coverage and complements targeted affinity-based platforms such as Olink.  By providing deeper access to the serum proteome, ENRICH-iST can reveal additional biological information that complements targeted protein quantification approaches.
Automatable, standardized workflow and higher reproducibility for reliable biological insights

Beyond plasma: Unlock challenging biofluids

ENRICH-iST is designed for researchers working across multiple sample types. The workflow supports challenging matrices such as synovial fluid and delivers deeper proteome coverage without the need for antibody-based depletion.

P2-iST Plasma – Redefining proteomics depth for plasma samples

P2-iST Plasma: combines proprietary P2 nanoparticle enrichment with the robust iST workflow to deliver up to 6-fold more protein identifications than neat-plasma preparation. Optimized specifically for human EDTA plasma11, the fast, standardized, and automation-ready workflow transforms raw plasma into LC-MS-ready peptides within hours. With reproducible throughput of up to 192 samples per day, it supports biomarker discovery in large-cohort studies.

Proteome depth that reveals disease biology

In a colorectal cancer pilot study (6 CRC patients vs. 6 healthy donors), P2-iST Plasma delivered an approximately 6-fold increase in plasma protein group identifications compared with neat plasma. Spike-in experiments with MART, IL-6, and IL-1β confirmed a linear quantitative response across a broad concentration range, from tens of pg/µL up to at least 12.5 ng/µL, supporting its suitability for targeted, biomarker-focused proteomics.

Uncovering biomarkers hidden in the plasma proteome

Applied to a biological cohort of P2-iST-enriched plasma samples (n=82) on the sensitive, high-performance PepSep Advanced column, the workflow enabled clear separation of patient groups by principal component analysis. Extracted ion chromatograms further revealed a lung cancer-specific intensity pattern for ITGA11, demonstrating that P2-iST Plasma preserves biologically meaningful, disease-associated signals for biomarker discovery.

In-Depth Characterization of the P2 Enrichment Workflow

Analysis of over 1,700 human EDTA plasma samples from 25+ sites worldwide identified a P2core proteome of 708 proteins consistently quantified across all samples, without data imputation. This core proteome remains stable across diverse collection methods and is notably enriched for vesicle-associated proteins, including 72 of the top 100 ExoCarta exosome markers — underscoring the robustness of P2 enrichment as a quality benchmark.

Automation-ready for scalable biofluid proteomics

"We tested several options for biomarker discovery on plasma samples, but many were long and tedious. With the novel ENRICH technology, we have a fast, efficient, and reliable enrichment workflow to get deeper into the proteome from human plasma and serum. We use the ENRICH-iST 96x kits and are delighted by the reproducibility and ease of the kit. We really enjoyed working with the PreOmics team, who was always there to support us. We would strongly recommend the kit to other plasma and serum users."

What do our customers say?

"At the Helsinki Proteomics Center, we have benchmarked essentially all major plasma depletion/enrichment strategies on the market. When depth matters, P2-iST consistently comes out on top. For us it has been a game changer – it puts robust, deep plasma proteomics into the hands of any lab without expensive proprietary hardware. The reproducibility and depth we see with P2-iST on our Orbitrap Astral platform makes it ideal for medium scale clinical and translational plasma studies."
