Characterization of proteins and biopharmaceuticals

Processing biologics such as monoclonal antibodies, purified proteins, drug substances and for multi-attribute monitoring (MAM) or host cell proteins (HCP) for analysis often requires optimized sample preparation procedures. Typical investigations include:
● Identification of post-translational modifications and their abundance or turn-over, peptide mapping for sequence validation (critical quality attribute analysis)
● Identification and quantification of HCP for quality assurance / quality control  and batch-to-batch analysis
● Biopharma CMC
          o   Quantification of specific residual proteins used in production e.g. specific enzymes used in production of small molecule drug or RNA which need to be below a defined limit.

The iST-BCT kit was developed to enable the robust processing of biological fluids, HCPs and therapeutic proteins such as monoclonal antibodies, and has been optimized to minimize artificial modifications (e.g. deamidation, oxidation) and increase the alkylation rate. The kit provides an all-in-one solution for processing samples in less than two hours and 30 minutes hands-on-time. HCP Analysis:

Plasma samples:

Non-depleted human plasma has a high protein concentration, commonly around 50 µg/µL. Mix up to 2 µL plasma/serum with 50 µL LYSE-BCT buffer.
