Tissues/FFPE

Tissues may be the starting point for a wide range of experimental investigations from discovery protein interaction studies, pharmacodynamic effects, through to large clinical trial biomarker measurements. For example looking at differential protein expression across a tissue section across a tumour, either from a recent biopsy or from a biobank FFPE slide. Homogenized tissues are compatible for processing with our iST and iST-NHS kits. .

The iST/iST-NHS kits are suitable for use in research and regulated environments. The kits contain all you need for your peptide or protein sample preparation and suitable for use by all members of your lab, no specialist experience is needed.

The flexible formats of single samples or racked in to 96 well holders allow the kits to be used manually or with a range of automation options. The cartridges can be processed by centrifugation or using positive pressure manifolds, again giving options for all labs. Dedicated resources are shown below and our technical support team would be happy to help with advice.

Please always ensure the total protein present is within the 1 – 100 µg range. Guidelines are given below dependent on the starting sample type.

Mammalian Tissue:

Mammalian tissue samples are more difficult to lyse and require some kind of mechanical force disruption, For example the PreOmics' BeatBox, in the presence of our LYSE or LYSE-NHS buffers. We also recommend addition of glass beads in combination with ultra sonication for most efficient tissue homogenization and cell lysis.

FFPE Tissue:

Processing of FFPE samples requires de-paraffinization of the FFPE punches/slides. In addition, de-paraffinized samples require a longer heat treatment for efficient de-crosslinking, increase the initial heating step from 10 to 60 min at 95°C.
